BackgroundProgrammedcelldeathligand1(PD-L1)alsoknownasclusterofdifferentiation(CD274)orB7homolog1(B7-H1)isatype1transmembraneproteininvolvedintheregulationofcellularandhumoralimmuneresponses.TheinteractionofPD-L1withitsreceptorPD-1providesbothstimulatoryandinhibitorysignalsinregulatingTcellactivationandtoleranceduringpregnancy,tissueallograft,autoimmunediseaseandmalignanttransformation.ThebindingofPD-L1toPD-1inducesapoptosisorexhaustioninactivatedTcells,andblockadeofthisinteractionhasbeenshowntoenhancetheantitumoractivityofTcells.PD-L1isfrequentlyoverexpressedinplacenta,andmanyhumantumourssuchasmelanoma,diffuselargeB-celllymphoma,andcarcinomasofthelung,bladder,breast,kidneyandcolon.HumanCD274,PD-L1.
