Thisisastandardispreparedbypoolingserumobtainedfromalargenumberofqualified(healthy,non-medicated)adultblooddonors.ThedeterminationofeachofthesubclasseswasrepeatedlydonebyELISAusingmonoclonalantiseratohumanIgAsubclassesandisolatedpolyclonalIgA1andIgA2fromnormalhumanserum,asprimarystandards.DifferentIgA1andIgA2paraproteinswerealsousedandtheresultsdidnotdifferfromthepolyclonalstandards.Toestimatetheinter-assayvariation,thesecondarystandardNOR-04andheprimarystandardswerescreenedseveraltimesonvariousdays.TheserumcontainsassignedvaluesforeachofthesubclassesofIgAandfortotalIgA:IgA1--2.00mg/ml.IgA2--0.20mg/ml.TotalIgA--2.20mg/ml.HumanStandardSerumNOR-04hasbeentestedandfoundnegativeforHbsAgandantibodiestoHIV(HTLVIII).Afterfiltration,accuratevolumesof0.25mlarefilledintovials.HumanStandardSerumNOR-04isintendedforuseasacalibratortopreparestandardcurvesinquantitativedeterminationsofthesubclassesofhumanIgA.

Storage:

Thisproductisshippedatambienttemperature.Afterarrivalstore,dividedintosmallaliquots,frozenatorbelow-20°C.Workingdilutionsmaybepreparedbyaddingtherequiredamountofphosphatebufferedsaline(PBS)containing0.05%Tweenand1%(w/v)bovineserumalbumin,shouldnotberefrozenandpreferablyusedwithin24hours.Repeatedthawingandfreezingshouldbeavoided.Ifaslightprecipitationoccursuponstorage,thisshouldberemovedbycentrifugationandwillnotaffecttheperformanceoftheproduct.At+4°C--1year.
