ThereactivityoftheantiserumisdirectedtotheFcsubunitoftheIgMmolecule,whichexpressesstrict(class)specificity.InimmunoelectrophoresisandrADIalinimmunodiffusionusingvariousantiserumconcentrationsagainstchickenserum,asingleprecipitinlinehasbeenobtainedwhichshowsareactionofidentitywiththeprecipitinlinesobtainedwiththepurifiedIgMusedasimmunogens.ItdoesnotreactwithIgG,IgG/FabfragmentsandIgAoranynon-Igproteininchickenserum,astestedbyimmunoelectrophoresisanddoubleradialimmunodiffusion.InimmunocytochemicalandimmunohistochemicalstainingofIgMatthecellularandsubcellularlevelofappropriatelytreatedcellandtissuesubstrates;todemonstratecirculatingIgMantibodiesinSEROdiagnosticmicroBIOLOGyandautoimmunediseases;toidentifyaspecificantigenusingareferenceantibodyofchickenoriginknowntobeoftheIgMisotypeinthemiddlelayeroftheindirecttestprocedure.Thisimmunoconjugateisnotpre-diluted.Theoptimumworkingdilutionofeachconjugateshouldbeestablishedbytitrationbeforebeingused.Excesslabelledantibodymustbeavoidedbecauseitmaycausehighunspecificbackgroundstainingandinterferewiththespecificsignal.
