Background:

Myeloperoxidase(MPO)isaglycoproteinpresentintheazurophil(primary)granulesofmyeloidcells,whichappearsinthemyeloblaststageofmyeloidcelldifferentiation.MPOishemostcommonfunctionalproteinofmyeloidcellsandisinvolvedintheinflammatoryresponse.Ithelpstokillmicrobesbybreakingdownperoxideinthepresenceofhalideions,contributingtothebactericidalfunctionofgranulocytes.TheprimarytranslationproductofMPOundergoesglycosylationwithproductionofthe89kDaheme-freeapopro-MPOformfollowedbyincorporationofhemeandconversionintotheenzymaticallyactivepro-MPOform.Subsequently,pro-MPObecomestargetedtoazurophilgranuleswherefinalprocessingoccurstoproducematuredimericMPOconsistingofthe59-64kDaMPOa-chainandthe14kDaMPOb-chain.PrecursorT-cellsaresurfaceCD3negativebutpositiveforcytoplasmaticCD3.AllmatureTcellsarebothcytoplasmicandsurfaceCD3positive.ThecombinedstainingforMPOandCD3allowsthedistinctionofcellsderivedformthemyeloidlineagethataregenerallyMPOpositive,fromimmatureandmatureTlymphocytesduringnormalandmalignanthematopoiesis.TheMPO-C2/CD3COMBI-ICreagentpermitstheidentificationandenumerationofnormalandmalignanthumanbloodandbonemarrowcellsusingflowcytometry.Resultsmustbeputwithinthecontextofotherdiagnostictestsaswellastheclinicalhistoryofthepatientbyacertifiedprofessionalbeforefinalinterpretation.Analysesperformedwiththisantibodyshouldbeparalleledbypositiveandnegativecontrols.Ifunexpectedresultsareobtainedwhichcannotbeattributedtodifferencesinlaboratoryprocedures,pleasecontactus.
