Membersofthetumornecrosisfactorreceptor(TNFR)familyareemergingasimportantmoleculesimpliCatedintheregulationofprolifeRation,differentiationandsurvivalofTandBlymphocytes.AmongthesereceptorsisCD27,thefunctionofwhichhasthusfaronlybeenstudiedintheHumansystem,whereitamplifiestheTcellprolifeRativeresponseinducedbyTCRtriggering.TheproteinencodedbythisgeneisamemberoftheTNF-receptorsuperfamily.ThisreceptorisrequiredforgeneRationandlong-termmaintenanceofTcellimmunity.ItbindstoligandCD70,andplaysakeyroleinregulatingB-cellactivationandimmunoglobulinsynthesis.ThisreceptortransducessignalsthatleadtotheactivationofNF-κBandMAPK8/JNK.AdaptorproteinsTRAF2andTRAF5havebeenshowntomediatethesignalingprocessofthisreceptor.CD27-bindingprotein(SIVA),apro-apoptoticprotein,canbindtothisreceptorandisthoughttoplayanimportantroleintheapoptosisinducedbythisreceptor.
