Thereactivityoftheantiserumisrestrictedtofibrinogen.InimmunoelectrophoresisandrADIalimmunodiffusion(Ouchterlony),usingvariousantiserumconcentrationsagainstnormaldogplasmaasingleprecipitinlineisobtainedwhichshowsareactionofidentitywiththeprecipitinlineobtainedwithpurifiedfibrinogen.Noreactionisobtainedwithanyotherplasmaproteincomponentorserum.However,theantiserummayalsoreactwithfibrinmonomers,circulatingfibrinopeptidesandfibrindegradationproducts.Asunlabelledprimaryorsecondaryantibodyreagentfortheindirectdetectionoffibrinogenindogcells,tissuesandbodyfluidsinimmunofluorescenceandimmunoenzymemethods;fortheproductionofimmunoconjugateswithaselectedMarker;toprepareinsolubleimmunoaffinityadsorbentsbycouplingtoanartificialcarrier;ascatchingordetectionreagentinnon-isotopicmethodologyandsolidphaseimmunochemistry(e.g.ELISA).Whenappliedinanycytochemicalorhistochemicalprocedureorsolidsphasecouplingtechnique,theoptimumconcentrationoftheIgGpreparationshouldalwaysbeestablishedbytitration.

Fibrinogen(clottingfactorI)isaheatlABIlebetaglycoproteinpresentinplasma.Itistheprecursoroffibrin,whichisthekeyproteinconstitutingthenetworkofthebloodclot.Thrombinconvertsfibrinogentofibrinbylimitedproteolysis.Fibrinmonomerspolymerizetofibrinwhichisstabilizedbycross-linking.Fibrinogenisisolatedfromfreshplasmaafterremovingprothrombin.Freund’scompleteadjuvantisusedinthefirststepoftheimmunizationprocedure.

Reconstitutethelyophilizedantiserumbyadding1mlsteriledistilledwater.Dilutionsmaybepreparedbyaddingphosphatebufferedsaline(PBS,pH7.2).Repeatedthawingandfreezingshouldbeavoided.Ifaslightprecipitationoccursuponstorage,thisshouldberemovedbycentrifugation.Itwillnotaffecttheperformanceoftheantiserum.

CrossReactivity:

TheantiSerumdoesnotcross-reactwithanyothercomponentofDogplasma.Inter-speciescrossreactivityisanormalfeatureofantibodiestoplasmaproteinssincetheyfrequentlyshareantigenicdeterminants.Cross-reactivityofthisantiSerumhasnotbeentestedindetail.
